A particular focus is on developing and applying in-cell NMR to quantify RNA conformational dynamics, including exchange between ground and excited states, directly in living human cells. Using enzymatic circularization of RNA to counteract intracellular degradation, and optimized relaxation-based data acquisition (selective excitation, SOFAST, and R1\u03c1 relaxation dispersion), the project will map how the cellular environment shapes the structural ensemble of biologically relevant RNA molecules.\nDuties\nThe work is primarily experimental (laboratory and NMR work) and includes:\nPreparation and purification of isotope-labeled RNA and DNA samples for NMR (in vitro transcription, large-scale production, and enzymatic circularization of RNA hairpins)\nIntroduction of nucleic acids into human cell lines (e.g., electroporation) and preparation of in-cell NMR samples, including controls for cell viability and leakage (supernatant control)\nCollection and optimization of solution NMR experiments for nucleic acids, including imino proton detection with selective excitation\/SOFAST and measurement of longitudinal relaxation (selT1)\nMeasurement of RNA dynamics using 1H and 15N R1\u03c1 relaxation dispersion to quantify populations, exchange rates, and chemical shifts of excited states, in vitro and in-cell\nQuantification of intracellular nucleic acid concentration and subcellular localization\nProcessing, curve fitting, and analysis of relaxation and dispersion data\nDocumentation and presentation of results\n\nWork is conducted in close collaboration with experimental and computationally-oriented researchers in the group.\nQualifications\nThe applicant should have a master's degree in structural biology, biophysics, biochemistry, chemistry, molecular biology, or a related field.
